Basic characteristics and structure of acridine esters

Acridinium Ester is a type of compound with a unique chemical structure, which contains the core structure of the acridine ring in its molecule. The substituent on the acridine ring can react with oxidants such as hydrogen peroxide (H ₂ O ₂) to generate unstable intermediates, which then release energy and convert it into visible light. This unique chemical property makes acridine ester an ideal chemiluminescent marker.

The basic principle of acridine ester chemiluminescence

1. Formation of excited state intermediates

Under alkaline conditions, acridine ester reacts with hydrogen peroxide to first produce an unstable ethylene oxide intermediate. This intermediate rapidly decomposes in a very short period of time, producing carbon dioxide (CO ₂) and an electronically excited N-methylacridone molecule. During this process, energy is stored in the excited state of N-methylacridone molecules.

2. Photon release

When the excited N-methylacridone molecule returns to its ground state, its stored energy is released in the form of photons, forming visible light. This process is similar to fluorescence, but the luminescence of acridine ester is more rapid and concentrated, with extremely high luminescence efficiency.

The advantages of acridine ester chemiluminescence technology

1. Low background illumination and high signal-to-noise ratio

The chemiluminescence reaction of acridine ester occurs under specific conditions with extremely low background noise, resulting in clearer detection signals and significantly improved signal-to-noise ratio. This feature is crucial for improving the sensitivity and accuracy of detection.

2. Rapid and concentrated luminescence reaction

The luminescence reaction speed of acridine ester is extremely fast, usually reaching the maximum luminescence intensity in about 0.4 seconds after adding the luminescence starter reagent, and the half-life is only about 0.9 seconds. This rapid and concentrated luminescent characteristic makes the detection process more efficient.

3. Easy to bind with proteins

Acridine esters can stably bind with biomolecules such as proteins, and the binding does not affect their photon yield. This characteristic has made acridine esters widely used in chemiluminescence immunoassay, achieving quantitative or qualitative detection of antigens or antigens by labeling them.

4. Stable markers

Acridine ester compounds can remain stable for a long time under appropriate storage conditions (such as 2-8 ℃), providing strong guarantees for the reliability and reproducibility of experimental results.
