The full name of PIPES buffer is piperazine-1,4-diethanesulfonic acid, a substance that can maintain the pH of the solution within a certain range. When a certain amount of acid and alkali is added to some solutions, it has the effect of hindering the pH change of the solution, which is called a buffering biological buffer. Such a solution is called a buffer solution. PIPES is an important member of Good’s buffer. The pH buffer range of PIPES is 6.1-7.5, insoluble in water, soluble in NaOH aqueous solution. PIPES is different from buffers containing two (2-hydroxyethyl) amino groups (such as Bis-tris, Bicine). It cannot form stable complexes with most metal ions, and is suitable for buffers in solution systems containing metal ions. According to existing research results, PIPES can be used to purify tubulin using phosphocellulose chromatography, to purify recombinant GTP-binding proteins ARF1 and ARF2 by gel filtration, and to use as a buffer to crystallize transketolase from E. coli. Since PIPES can form free radicals, it is not suitable for redox systems. In cation exchange chromatography, a low concentration of PIPES buffer should be used, because PIPES has a relatively large ionic strength and its pKa value is concentration-dependent.
Preparation method of PIPES solution (about 1-2L) with different pH value
1. With 0.1M solution (A)
(1) NaOH/deionized water=4g/400ml …①

2. With 0.1M NaOH solution (B): NaOH 4g/ deionized water 1,000ml
